The 16S rRNA gene approach was applied to investigate the diversity of Planctomycetes in latitudinal surface seawater of the Western Pacific Ocean. The results revealed that the Pirellula-Rhodopirellula-Blastopirellula clade dominated the Planctomycetes community at all surface seawater sites while the minority genera Gemmata and Planctomyces were only found at sites H5 and H2 respectively. Although the clone frequency of the PRB clade seemed stable (between 83.3% and 94.1%) for all surface seawater sites, the retrieved Pirellula-Rhodopirellula-Blastopirellula clade presented unexpected diversity. Interestingly, low latitude seawater appeared to have higher diversity than mid-latitudes. integral-LIBSHUFF software analysis revealed significantly different diversity patterns between in latitudinal surface seawater and in the sediment of South China Sea station M2896. Our data suggested that different hydrological and geographic features contributed to the shift of Planctomycetes diversity in marine environments. This is, to our knowledge, the first systematic assessment of Planctomycetes in latitudinal surface seawater of the open sea and the first comparison of diversity pattern between surface seawater and sediments and has broadened our understanding of Planctomycetes diversity in marine environments.  相似文献

Most of the ambient dissolved organic carbon (DOC) is refractory to microbial degradation; bacteria can consume a minor but variable part of the DOC pool over periods of hours and days. It is important to increase our knowledge of the dynamics of the biodegradable fraction of DOC (BDOC) to understand the global carbon budget.Several methods for determining BDOC have been developed; however, the problem of most of them is the time (days/weeks) required for the colonization and/or determination.In this paper, we describe the application to seawater of a plug-flow bioreactor to measure BDOC within 3–4 h. The bioreactor was built following Søndergaard and Worm [Søndergaard, M., Worm, J., 2001. Measurement of biodegradable dissolved organic carbon (BDOC) in lake water with a bioreactor. Water Res. 35, 2505-2513.] protocols for the measurement of BDOC in lake water. We analyzed BDOC on samples collected in the Gulf of Trieste during autumn–winter and summer 2003–2004. BDOC concentrations varied from 8 to 24 μM and represented from 10.3% to 25.5% of the total DOC. To evaluate the effectiveness of this method, we compared bioreactor BDOC measurement with data obtained from batch cultures. The results indicate that BDOC in coastal seawater can be measured rapidly and reliably with this bioreactor.  相似文献

Dissolved organic matter (DOM) in seawater can be defined as the fraction of organic matter that passes through a filter of sub micron pore size. In this study, we have examined the effect of DOM of deep seawater (DSW) from Pacific Ocean on platelet aggregation and atherosclerosis progression. DSW was passed through a series of filters and then through an Octadecyl C18 filter; the retained substance in ethanol was designated as C18 extractable DOM (C18-DOM). Our studies showed that C18-DOM treatment inhibited platelet aggregation, P-selectin expression and activity of COX-1 significantly. C18-DOM increased the expression of anti-atherogenic molecule namely heme oxygenase-1 in endothelial cells and all these data showed that C18-DOM is exhibiting aspirin-like effects. Moreover our in vivo studies showed that C18-DOM feeding slowed remarkably the progression of atherosclerosis. Our study demonstrated a novel biological effect of oceanic DOM, which has several important implications, including a possible therapeutic strategy for atherosclerosis.  相似文献

The sea anemone Anthopleura aureoradiata (Carlgren), which harbours symbiotic dinoflagellates (zooxanthellae), is abundant on mudflats and rocky shores around New Zealand. We measured the potential for particulate nitrogen uptake from sediment by A. aureoradiata and the subsequent consequences of this uptake on the nitrogen status of its zooxanthellae. Sediment was rinsed, labelled with (15NH4)2SO4, and provided to anemones at low (0.23 g ml− 1) and high (1.33 g ml− 1) sediment loads for 6 h. Both anemone tissues and zooxanthellae became enriched with 15N. Enrichment of anemone tissues was similar at both high and low sediment loads, but the zooxanthellae became more enriched at the lower load. This was presumably because the uptake of ammonium, arising from host catabolism, by zooxanthellae is light driven and because the anemones at the lower load were able to extend their tentacles into the light while those at the higher load were not. The influence of sediment uptake on the nitrogen status of the zooxanthellae was determined by measuring the extent to which 20 μM NH4+ enhanced the rate of zooxanthellar dark carbon fixation above that seen in filtered seawater (FSW) alone; the ammonium enhancement ratio (AER) was expressed as [dark NH4+ rate/dark FSW rate], where ‘rate’ refers to C fixation and a ratio of 1.0 or less indicates nitrogen sufficiency. When anemones were starved with and without rinsed sediment in nitrogen-free artificial seawater for 8 weeks, zooxanthellar nitrogen deficiency became apparent at 2-4 weeks and reached similar levels in both treatments (AER = ~ 2). In contrast, anemones fed 5 times per week for 8 weeks with Artemia nauplii were nitrogen sufficient (AER = 1.03). In the field, zooxanthellae from mudflat anemones were largely nitrogen sufficient (AER = 1.26), while nitrogen deficient zooxanthellae were present in anemones from a rocky intertidal site (AER = 2.93). These results suggest that, while there was evidence for particulate nitrogen uptake, dissolved inorganic nitrogen (especially ammonium) in interstitial pore water may be a more important source of nitrogen for the zooxanthellae in mudflat anemones, and may explain the marked difference in nitrogen status between the mudflat and rocky shore populations.  相似文献

The effects of hypothermal (22–16?°C) and hyperthermal (22–28?°C) stress on the immune system responses of the shrimp Litopenaeus vannamei cultured in either freshwater or seawater were measured and compared. The following immune system indicators were measured for comparison: total hemocyte count (THC), activity of phenoloxidase (PO), nitric oxide synthase (NOS), superoxidase (SOD), and malondialdehyde (MDA) content. Thermal stress significantly decreased THC in both freshwater and seawater shrimp within 6–12?h (P?0.05). After hypothermal stress, all shrimp had a significantly lower THC level than their prechallenge levels (P?0.05). Under both types of thermal stress, the activity of PO, NOS, and SOD first increased and then decreased. After 48?h of thermal stress, shrimp PO and NOS activity decreased in both freshwater and seawater. After 48?h of thermal stress, the reduction in the SOD activity in the hemolymph of freshwater shrimp was greater than that in seawater shrimp. During exposure to stress, the MDA content in freshwater shrimp was significantly higher than in seawater shrimp, which demonstrated that lipids in freshwater shrimp were more susceptible to peroxidation than those in seawater shrimp, particularly at low temperatures. Large temperature fluctuations, particularly sudden cooling, should be avoided when rearing L. vannamei because of high rates of lipid peroxidation and decreased immunity. These effects are more marked in freshwater than in seawater.  相似文献

种植抗盐耐海水经济作物是合理开发利用沿海滩涂和海水资源、培育耐盐植物的有效措施之一。该研究以不同产地菊芋(Helianthus tuberosus)为试验材料研究了海水处理对菊芋幼苗生长、光合作用和叶绿素荧光特性的影响。结果表明:1)随着海水浓度的增加,各地菊芋幼苗根、地上部的鲜重和干重均降低,但盐城菊芋幼苗地上部和根鲜重及干重降低的幅度小于武威和烟台菊芋幼苗。2)随着海水浓度的增加,各地菊芋叶片的净光合速率(Pn)、蒸腾速率(Tr)、水分利用效率(WUE)、气孔导度(Gs)和气孔限制值(Ls)均降低,而细胞间隙CO2浓度(Ci)增大,且在各处理下盐城菊芋较武威和烟台菊芋叶片PnTr大,而盐城菊芋在15%海水处理下,WUECi较武威及烟台菊芋低,GsLs较武威和烟台菊芋高。3)随着海水浓度的增加,各地菊芋叶片的初始荧光(Fo)、最大荧光(Fm)、可变荧光(Fv)、恒态荧光(Fs)、恒态荧光与初始荧光差值(ΔFo)、PSⅡ潜在光化学效率(Fv/Fo)和PSⅡ最大光化学效率(Fv/Fm)均降低,且在各处理下盐城菊芋较武威和烟台菊芋叶片FmFvFsFv/Fo大。随着海水浓度的增加,盐城菊芋叶片电子传递速率(ETR)、光化学荧光猝灭系数(qP)和光化学速率变化幅度不大,武威和烟台菊芋在30%海水处理下显著下降,较盐城菊芋小,各处理下菊芋叶片的PSⅡ实际光化学效率(ΦPSⅡ)变化也不一致,盐城和武威菊芋叶片的ΦPSⅡ在15%海水处理较0和30%海水处理下大,而烟台菊芋叶片的ΦPSⅡ在0海水处理下最大。4)随着海水浓度的增加,各地菊芋叶片的叶绿素a(Chla)含量均降低,而叶绿素b(Chlb)含量没显著差异,且在各处理下盐城菊芋较武威和烟台菊芋叶片Chla含量大,武威和盐城菊芋叶片的Chla/Chlb比值均降低,烟台菊芋叶片的Chla/Chlb比值在0和15%海水处理下没差异。可见海水处理对菊芋幼苗生长发育、光合作用和叶绿素荧光特性均有影响,随海水浓度的增加,其效应越明显,但对各来源地菊芋幼苗的影响不一致,盐城菊芋幼苗表现为更耐海水。  相似文献

Increases in size of Colpomenia peregrina (Sauv.) Hamel microthalli were determined at six different combinations of salinity and temperature under laboratory conditions. Microthalli grown at low salinity (15%) did not have a growth rate significantly different from microthalli grown at high salinity (30%). However, more macrothalli were produced in higher salinity media. Fewer macrothalli were produced at lower temperatures. Growth of C. peregrina microthalli was also determined at six different combinations of daylength and temperature. Microthalli under 15:9 h (LD cycle) did not have a growth rate significantly different from microthalli under 9:15 h LD. However, more macrothalli were produced under long day conditions. Of the three different temperatures utilized (5, 13 and 20° C) only 5° C produced significantly different growth rates. Low temperature reduced growth to such an extent that macrothalli of a size that could be recognized in the field took approximately two months to produce in culture. These results may explain the seasonal presence/absence of the delophycean phase of C. peregrina in the field. Zoospore survivorship and macrothallus production of C. peregrina in culture indicated that both PES and an artificial medium were suitable for the laboratory cultivation of this plant.  相似文献

Results of a chemical and geochemical study of the Oslofjord system are presented to explore the possibility of qualitatively identifying a eutrophication gradient. Nutrient concentration and dissolved oxygen may indicate such a gradient in areas of similar hydrographies. Uncertainties due to minor differences in hydrography may be resolved by considering sedimentary organic carbon, C:N ratios and the accumulation of mineralisation products in the sediment pore water. The reliability of the sedimentary organic matter and the pore water composition as gradient indicators can be impaired by input of terrestrial organics, dispersal of mineralisation products by the benthic fauna or the apparent high levels of organic carbon and pore water nutrients in the absence of the benthic fauna under anoxic conditions.  相似文献

海水养殖尾水中总氮、总磷超标是引起沿海水体富营养化的主要原因,为研究碱蓬浮床对海水养殖尾水中氮磷的去除效果,该研究设计加入碱蓬(Suaeda salsa)浮床和不加浮床的两组对比实验,通过比较修复前后碱蓬株高、生物量、含水率、根长以及各部位氮、磷的含量变化,以及水体中总氮(TN)和总磷(TP)的去除效果,探究浮床中碱蓬对总氮和总磷的吸收及其生长特性,验证碱蓬浮床对海水养殖废水中氮、磷等的去除能力。结果表明:浮床中碱蓬株高、鲜重、干重、含水率、根长较修复前均有显著增加,说明浮床中盐生植物碱蓬能够适应含海水养殖尾水水培环境;经碱蓬浮床修复,水体中总氮、总磷均明显下降,其中碱蓬对海水养殖尾水中的总氮总磷去除贡献率分别为16.10%和78.15%,浮床中碱蓬会在叶片和根系中积累氮磷。  相似文献
